Characterization of Polysaccharide-based Nanoparticles for Targeted Cancer Immunotherapy

I am proud to present my work on a pilot study with the University of Pittsburgh Dr. Song Li Lab. I was given the opportunity to manage a pilot study to assess the viability of polysaccharide-based polymers for drug delivery of therapeutic mRNA and immunotherapeutics to tumor tissue. I was tasked with synthesizing a chemical library based on 16 novel polymer backbones over a 3 month period. By brainstorming more backbones and continually synthesizing chemicals, I was able to formulate a chemical library of 162 novel nanoparticles, all capable of aqueous mixing with one another, in order to tailor properties. In order to streamline and expedite discovery, only 92 of the list of 162 nanoparticles were synthesized and formulated for testing. I was responsible for synthesis and storage of precursors as well as ensuring products were pure. I was expected to adhere to responsible conduct of research policies, maintain an orderly workspace, and keep a detailed lab book. I was encouraged to collaborate with neighboring labs and lab members. For testing, each nanoparticle was loaded with a combination therapy consisting of therapeutic mRNA and an immunotherapeutic. Their structure, stability, toxicity, and effectiveness were all assessed using various assays and cell-culture methods (Panc02 model). With the help of extremely generous senior lab members, I was able to assess these novel treatments in-vivo as well (mouse models). Some promising data has already been found, with more discovery on the horizon! A special thanks to the Dr. Song Li lab and all of its current and former lab members. They made me truly believe I could not only be the scientist I always dreamed, but a great one.
